Melatonin is a natural neurohormone produced in humans by the pineal gland. A favourite of travellers, it governs the body’s circadian rhythms, helping the body ease into restful sleep and is also a potent antioxidant. However, in other contexts and higher dosages it has been found to have strong anti-estrogenic effects (after 4 months of HD usage) and even stronger actions to ER Stress which if sustained can initiate programmed cell death (PCD) to tumor cells via UPR, Melatonin at high dosages has also been found to enhances the cytotoxicity of targeted anti-tumor drugs.

Melatonin and endoplasmic reticulum stress: relation to autophagy and apoptosis.

The hormone melatonin produces a number of health benefits in terms of your immune system. It’s a powerful antioxidant and free radical scavenger that helps combat inflammation. In many tumors, cancer cells take-up LA via a cAMP-dependent transport mechanism and metabolise it to the mitogen 13-hydroxyoctadecadienoic acid (13-HODE) by the enzyme 15-lipoxygenase-1 , the activity of which is up-regulated by activation of epidermal growth factor (EGF) and insulin-like growth factor-1 (IGF-1) receptors . Peer-reviewed and published research has shown Melatonin offers particularly strong protection against reproductive tumors. Cells throughout your body — even tumor cells — have melatonin receptors. So when melatonin makes its nightly rounds, cell division slows. When this hormone latches onto a breast tumor cell, it has been found to counteract estrogen’s tendency to stimulate cell growth.

In fact, melatonin has a calming effect on several reproductive hormones, which may explain why it seems to protect against sex hormone-driven tumors, including ovarian, endometrial, breast, prostate and testicular tumor . But melatonin’s anti-tumor effects don’t stop there. Melatonin is derived almost entirely from the pineal gland, as shown by the fact that undetectable melatonin levels are found [8]. After its release, melatonin is bound to albumin and reaches tissues within a very short period [9]. The melatonin molecule possesses lipophilic and hydrophilic properties that permit its transfer into many tissues and fluids. Depending upon the site of production and target organ, melatonin can act as a hormone, immuno-modulator or as a biological modifier [10].

Melatonin has been identified as a natural chronobiotic substance with immune-enhancing properties as well as the ability to reduce oxidative stress. Thus, melatonin could potentially be used as a therapeutic substance for the prevention or arrest of neoplastic growth [11]. The concentrations of melatonin in ovarian follicles tend to increase with follicular growth [12]. It is possible that melatonin acts as an antioxidant [13]. Furthermore, the anti-apoptotic effect of melatonin has been observed in a variety of cell types [14].  And there are indications that melatonin can help prevent and treat oxidative damage as well as suppress specific inflammatory factors [15].

 

Tumor Therapy Adjuvant and Immune Regulator

Emerging research suggests that melatonin has anti-tumor properties—that is, it has the ability to prevent tumors from occurring, or to induce the tumor cell death if it does occur. This has been attributed to melatonin’s antioxidant, anti-inflammatory, anti-proliferative, and hormone-modulating properties.

Melatonin’s ability to interfere with tumor cell multiplication and growth (“proliferation“), as well as inducing tumor cell death (“apoptosis”), has been documented in tumor patients.

It has been successfully used in individuals with advanced stage tumors undergoing conventional antitumor therapy, by either slowing disease progression and/or decreasing treatment side effects. 53-60 In a review of 8 randomised, controlled clinical trials evaluating the benefits of melatonin as an adjuvant therapy for tumor patients with solid tumors undergoing chemotherapy or radiation therapy. Researchers found that concurrent use of 20 mg of melatonin once daily versus conventional treatment alone, improved the rate of complete or partial remission by nearly 50%, increased the one-year survival rate by 45%, and decreased the devastating side effects of conventional therapy such as low platelet count, neuropathy, and fatigue by 89%, 83%, and 65% respectively. 59 What’s more, these effects were consistent across different types of tumor and there were no adverse events reported.

Melatonin’s anti-tumor properties can also be attributed to its effect on your immune system. Laboratory studies revealed that melatonin can activate T-helper cells, which triggers other immune cells in order to help kill off foreign invaders or pathogens.  Additionally, melatonin stimulates natural killer cell, monocyte, and macrophage synthesis, and has been found to facilitate healthy cell-to-cell communication, which enhances the body’s appropriate immune system response to foreign invaders.

Based on available evidence, leading experts suggest that patients with tumors and particularly metastatic solid tumors, might benefit from melatonin use, potentially leading to improved therapeutic outcomes.   Certainly, more research is warranted.

Melatonin not only inhibits breast tumor cell growth, but also is capable of inhibiting angiogenesis, tumor cell invasion and telomerase activity. Interestingly this hormone is able to induce apoptosis through the suppression or induction of a wide range of signaling pathways. Moreover, it seems that the concomitant administration of melatonin with other conventional chemotherapy agents had beneficial effects for patients with breast tumor , by alleviating unfavorable effects of those agents and enhancing their efficacy.

Suggested Use:

Due to the potentially sedative nature of melatonin, start with 1/2 or 1/3 capsule emptied into water or less and assess your personal response and gradually build up over a few days. If you do not experience a deep restful sleep, your dose may have been too low.  Do not exceed the suggested use unless recommended by your physician. Mel Magic taken too close to bedtime can be stimulating due to the Green Tea and the Salvia . So try to take it 2 hours before bedtime or possibly an hour after the evening meal.

Despite melatonin’s antioxidant properties, it has demonstrated a tendency to stimulate inflammation in people with certain autoimmune disorders. A study published in the October 2007 issue of the “British Journal of Clinical Pharmacology” demonstrated that rheumatoid arthritis slightly worsened in study subjects who took melatonin [source] .
