It was almost half a century before the first human tumor virus, Epstein-Barr virus (EBV), was identified in 1964. Subsequently, several human tumor viruses have been identified including Kaposi sarcoma associated herpesvirus (KSHV), human Papillomaviruses (HPV), Hepatitis B virus (HBV), Hepatitis C virus (HCV), Human T lymphotropic virus (HTLV-1) and recently identified Merkel cell Polyomavirus (MCPyV).
Tumor viruses are sub-categorized as either DNA viruses, which include EBV, KSHV, HPV, HBV, and MCPyV, or RNA viruses such as HCV and HTLV-1. Tumor-viruses induce oncogenesis through manipulating an array of different cellular pathways.
These viruses initiate a series of cellular events, which lead to immortalization and proliferation of the infected cells by disrupting the mitotic checkpoint upon infection of the host cell. This is often accomplished by functional inhibition or proteasomal degradation of many tumor suppressor proteins by virally encoded gene products.
The virally infected cells can either be eliminated via cell-mediated apoptosis or persist in a state of chronic infection. Importantly, the chronic persistence of infection by tumor viruses can lead to oncogenesis which is an uncontrolled proliferation of growth.
Tumor virus encoded oncoproteins deregulate components of the cell cycle and apoptotic pathways. viral-oncoproteins (red ovals) interact with and manipulate normal functions of numerous cellular important proteins (black) to hijack entire cell cycle and apoptotic machineries.
VBLOCK™ is a broad-based organic nutraceutical formula containing herbal extracts and other nutrients which have been shown in previous studies to provide antiviral support and may reduce inflammation. VBLOCK™ attempts to block the pathways which are used for replication and infection of immune system B cells. VBLOCK™ has not been proven in this respect though it’s individual ingredients have been shown to provide antiviral efficacy.
Research in virology has provided meaningful links to mammalian checkpoint control elements with the characterisation of growth‑promoting proto‑oncogenes encoding c‑Myc, Mdm2, Cyclins A, D1 and G1, and opposing tumor suppressor proteins, such as p53, pRb, p16INK4A and p21, which are commonly dysregulated in cell mutations. Cyclin D is a member of the cyclin protein family that is involved in regulating cell cycle progression. The synthesis of cyclin D is initiated during G1 and drives the G1/S phase transition.
Cyclin D is one of the major cyclins produced in terms of its functional importance. It interacts with four Cdks: Cdk2, 4, 5, and 6. In proliferating cells, cyclin D-Cdk4/6 complex accumulation is of great importance for cell cycle progression. Namely, cyclin D-Cdk4/6 complex partially phosphorylates retinoblastoma tumor suppressor protein (Rb), whose inhibition can induce expression of some genes (for example: cyclin E) important for S phase progression.
Natural compounds and its derivatives have been widely used into clinical treatment due to its innovative structure, high biological activity and low side effects. However, its complicated mechanism is difficult to be elucidated, which limit its application. Increasing evidence has revealed that some natural compounds exhibit anticancer properties through epigenetic regulation. During its antitumor action, natural compounds have been shown to alter expression profiles of certain miRNAs by many mechanisms. [source]

Recent advances in genetic engineering, functional genomics and clinical oncology converged in identifying cyclin genes and CDK’s as a pivotal components of a commanding cyclin/Mdm2/p53 axis and a strategic locus for re‑establishing cell cycle control by means of therapeutic gene transfer.
